In C of Ammonia

Leaf attached to seedling plant which was dug up for the purpose was immersed in Ammonia 4 per 1000 10.30 am Sep 8th. Examined Sep 9th 11.30; the leaf looks perfectly healthy, & is now washed with distilled water & left under bell-glass

There is no aggregation of chlorophyll which looks healthy; but in the epidermis cells especially the upper epidermis there are are large yellowish highly refracting spheres in each cell. These are easily soluble in alcohol and which leaves a shrunk remnant looking like dead protoplasm

Sep 10th 10.30 am leaf crumpled & withered at edges

(Oct 15th Leaf in C. of amm. for 26 hr. 4 to 1000 —no aggregation Chl. grain in part well preserved, in parts become granular in epidermis cells transparent, almost colourless, very slightly greenish-brown.— In sol. of 7 to 1000 for 27 hr no aggregation. The transparent spheres in epidermis disappear instantly when acetic acid added under cover glass leaving very slight granular residue.
